# Metformin Findings: Increases Healthspan Without Requirement Function and Lung Function ## Signal Metformin increases healthspan without a requirement for function of the insulin signaling pathway. reports: Median survival for animals on 0 mM and 50 mM metformin is 24 and 31 days, respectively, and the survival curves are significantly different by the Log-rank test ( P = 0.0014). We found similar median survival extension in a single repeat of this experiment (pooled data for the two trials show an approximate 36% median lifespan increase and significantly different survival curves ( P Table S1C ). [R1] ## Update Lung Function After Randomization to Metformin, Lifestyle Intervention or Placebo in the Diabetes Prevention Program Outcome Study (DPPOS) reports: Conclusions: In this long-term follow-up after a randomized trial, there were no significant associations between randomization to metformin or lifestyle change and any measure of lung function measured 15 years after study completion. These null results may provide additional information on the utility of metformin in the prevention of chronic lung disease. [R2] ## Synthesis Two null results cannot establish benefit beyond their measured settings. They are independent findings in distinct populations and endpoints, not a coordinated trial program, and cannot be pooled or generalized. Therefore, any proposed metformin benefit must name its population and endpoint; this evidence does not prove general inefficacy. [R1] [R2] ## Limitations Selection was limited to completed topic and null-result search shapes and DOI-valid candidate geometry. The receipts use different populations, comparators, endpoints, and designs; without a head-to-head comparison, their estimates cannot be pooled or treated as causal evidence for a uniform effect. A receipt identified as an abstract record carries lower evidentiary weight than a full peer-reviewed article. [R1] [R2] ## Falsifier For Metformin increases healthspan without a requirement for function of the insulin signaling pathway., a preregistered randomized study in its stated population showing a reproducible benefit on Increases Healthspan Without Requirement Function would overturn the first boundary. For Lung Function After Randomization to Metformin, Lifestyle Intervention or Placebo in the Diabetes Prevention Program Outcome Study (DPPOS), long-term randomized follow-up showing a reproducible benefit on Lung Function would overturn the second.
