# Alpha memo: resveratrol exercise context boundary
**One-sentence alpha:** Resveratrol plus exercise was feasible to combine in a small pilot RCT of functionally limited older adults, but a separate trial suggests resveratrol may not add to exercise's metabolic/inflammatory signal in aged skeletal muscle.
**Receipt 1:** Harper et al. (2021), "Resveratrol and exercise combined to treat functional limitations in late life: A pilot randomized controlled trial," reports that a three-arm pilot RCT (N=60, ~72-year-old adults with functional limitations) of 12 weeks of center-based walking plus resistance training combined with 0/500/1000 mg/day resveratrol was framed as designed to evaluate safety and feasibility, with physical function and muscle mitochondrial outcomes as secondary measures.
**Receipt 2:** Olesen et al. (2014), "Exercise training, but not resveratrol, improves metabolic and inflammatory status in skeletal muscle of aged men," reports that in healthy inactive men aged 60–72, 8 weeks of high-intensity exercise training increased skeletal muscle PGC-1α mRNA (~1.5-fold) and markers of oxidative capacity and NF-κB signaling, while 250 mg/day resveratrol alone showed no additive effect to exercise on these skeletal muscle endpoints.
**Why this is surprising:** Receipt 1 made resveratrol-with-exercise a plausible combined intervention worth testing in late-life functional limitation, yet Receipt 2 suggests the same combination yields no additive metabolic/inflammatory effect in aged skeletal muscle, hinting at a split rather than a uniform benefit.
**Caveats/falsifiers:**
- Receipt 2 used a different modality (high-intensity training, endurance test), lower resveratrol dose (250 mg), 8-week duration, and healthy inactive men versus Receipt 1's functionally limited older adults at 500/1000 mg over 12 weeks, so the contrast is confounded across multiple axes (dose, duration, baseline status, modality, sample size) and any single-moderator explanation is tentative.
- Sample sizes are small in both trials (Receipt 1 N=60, Receipt 2 healthy inactive aged men), so a decisive falsifier would be an adequately powered RCT in functionally limited older adults directly testing whether resveratrol adds to exercise on both functional and muscle mitochondrial endpoints.
