# Alpha memo: exercise resveratrol muscle context boundary
**One-sentence alpha:** Co-administering resveratrol with exercise may improve selected metabolic and muscle molecular readouts while leaving body-weight loss and intramyocellular lipids unchanged, suggesting endpoint- and context-dependent effects rather than uniform benefit.
**Receipt 1:** *The Effect of Periodic Exercise and Resveratrol Supplementation on the Expression of Pparg Coactivator-1 Alpha and Pyruvate Dehydrogenase Kinase Genes in Gastrocnemius Muscle of Old Rats With Type 2 Diabetes* (2019) — aimed to assess PGC-1α and PDK4 gene expression in gastrocnemius muscle of old type-2-diabetic rats across exercise, supplement, and combined groups (a planned factorial design reporting gene-expression endpoints, not a confirmed clinical benefit).
**Receipt 2:** *Early potential effects of resveratrol supplementation on skeletal muscle adaptation involved in exercise-induced weight loss in obese mice* (2018) — reported that combined exercise plus resveratrol over four weeks in high-fat-diet obese mice exerted no additional effects on body-weight loss, significantly improved whole-body glucose and lipid homeostasis, decreased intrahepatic lipid content without affecting intramyocellular lipid content, and increased mtDNA, cytochrome c, and PGC-1α-related signaling.
**Why this is surprising:** Receipt 1 framed exercise+resveratrol as a combined intervention worth testing for skeletal-muscle gene-expression synergy, but Receipt 2 shows the combination does not add to weight loss and is silent on many gastrocnemius endpoints, indicating the same pairing can be neutral on some outputs while active on others.
**Caveats/falsifiers:**
- Receipt 1 is a 42-rat factorial design in streptozotocin-induced diabetic rats (PGC-1α/PDK4 in gastrocnemius) and its combined-arm gene-expression result is not observable here; Receipt 2 used obese mice on high-fat diet over 4 weeks — populations, species, diabetes status, durations, and endpoints differ on multiple axes, so any species- or context-moderator explanation is tentative and confounded.
- Receipt 1 (2019) is co-temporal mechanistic context rather than a direct replication of Receipt 2 (2018); no clinical, dosing, or supplementation recommendation follows from these two receipts, and dose-equivalent scaling across them is not established.
- A decisive future falsifier would be a factorial rodent study in matched population, species, dose, and duration that reports combined exercise+resveratrol significantly reducing intramyocellular lipid or adding to body-weight loss contrary to Receipt 2's null/no-additive pattern.
