# Alpha memo: Resveratrol + exercise may lose or invert the liver/cardiovascular signal between mature-mouse models and older adults
**One-sentence alpha:** Resveratrol added to exercise appears to support liver anti-aging readouts in mature SAMP8 mice, but in older adults a 12-week combined intervention failed to translate into a clean TMAO/CVD-risk benefit, suggesting context-dependent decoupling.
**Receipt 1:** Combination resveratrol with exercise training has anti-aging function only at mature stage SAMP8 mice liver — in 6-month-old SAMP8 mice, combined habitual exercise + resveratrol intake yielded lower adipocyte accumulation, collagen, and apoptosis in liver cross-sections versus 3-month-old mice, and was associated with modulated PI3K/Akt, Bad, Bcl-2, IL-6/STAT3, and ERK5/FGF2/MMP2 protein markers.
**Receipt 2:** Effects of resveratrol on changes in trimethylamine-N-oxide and circulating cardiovascular factors following exercise training among older adults — in 41 adults aged ~72 years randomized to placebo, 500 mg/day, or 1000 mg/day resveratrol alongside 12 weeks of twice-weekly 80-minute supervised exercise, the abstract frames human experimental evidence for combined resveratrol + training effects on TMAO and related cardiovascular metabolites as still unresolved.
**Why this is surprising:** Receipt 1 made plausible that pairing resveratrol with exercise is a coherent, organism-level positive signal for aging liver tissue; Receipt 2 updates this by showing the same combination, when moved into older humans, does not yet demonstrate a clear dose-responsive TMAO or CVD-metabolite benefit.
**Caveats/falsifiers:**
- Receipt 1 is in SAMP8 mice at a single mature age window with histological and western-blot readouts; Receipt 2 is a small (n=41), 12-week, two-dose trial in adults ~72 years old with high baseline CVD risk — neither sex-, species-, nor dose-equivalent comparability is established.
- A future multi-dose RCT in adults using matched training volume and standardized TMAO/cardiometabolic endpoints, and showing a clear dose-responsive TMAO reduction with resveratrol + exercise, would falsify the suggested context-dependent decoupling.
