# Alpha memo: nicotinamide riboside exercise performance context boundary
**One-sentence alpha:** Nicotinamide riboside supplementation suggests context-dependent exercise effects that may split by age/baseline status rather than acting as a uniform ergogenic.
**Receipt 1:** The NAD+ precursor nicotinamide riboside decreases exercise performance in rats (2016) — chronic 21-day NR at 300 mg/kg/day via gavage in 18 Wistar rats showed a tendency toward worse physical performance on an incremental swimming test versus saline vehicle.
**Receipt 2:** Acute nicotinamide riboside supplementation improves redox homeostasis and exercise performance in old individuals (2020) — acute NR in 12 old men (with 12 young men for comparison) increased NAD(P)H levels, decreased oxidative stress, and improved physical performance in the old group, who started with lower erythrocyte NAD(P)H and higher oxidative-stress markers.
**Why this is surprising:** Receipt 1 made plausible the expectation that an NAD+ precursor would robustly enhance exercise across settings, yet Receipt 2 reframes the signal as bounded by deficiency/age context rather than a uniform benefit.
**Caveats/falsifiers:**
- Receipt 1 used 300 mg/kg/day for 21 days in young male Wistar rats (n=9/group, swimming test); Receipt 2 used a single acute dose in 12 young vs 12 old men — species, dose, duration, baseline status, and sample size all differ, so any age/deficiency moderator hypothesis is tentative and confounded by these other axes; an equivalent cross-species, dose-matched, chronic-vs-acute comparison that abolishes the old-group gain would decisively falsify the context-dependence story.
