# Alpha memo: Split response of nicotinamide riboside on exercise performance by age
**One-sentence alpha:** Nicotinamide riboside (NR) supplementation may shift from performance-impairing in young rats to performance-improving in older humans when baseline NAD(P)H status is low.
**Receipt 1:** "The NAD+ precursor nicotinamide riboside decreases exercise performance in rats" — In a 21-day gavage protocol at 300 mg/kg/day in Wistar rats, the NR group showed a tendency towards worse swimming performance than saline controls.
**Receipt 2:** "Acute nicotinamide riboside supplementation improves redox homeostasis and exercise performance in old individuals: a double-blind cross-over study" — In young and old men given a single acute NR dose 2 hours before testing, only old participants (who had lower baseline erythrocyte NAD(P)H and higher oxidative stress) showed increased NAD(P)H levels, decreased oxidative stress, and improved physical performance after NR.
**Why this is surprising:** The same molecule and class of effect (NR + exercise) diverges in opposite directions across species, age, and dosing regimen, suggesting baseline redox deficit — not just NR exposure — gates whether the signal is positive.
**Caveats/falsifiers:**
